BBI-825 is a novel, orally available, selective small molecule inhibitor of ribonucleotide reductase (RNR), the rate-limiting enzyme responsible for the cellular production of deoxynucleotide triphosphates (dNTPs), which are essential building blocks for DNA synthesis and repair. Developed by Boundless Bio using its proprietary Spyglass platform, BBI-825 is designed to target extrachromosomal DNA (ecDNA)-enabled cancers that rely on oncogene amplification for growth and resistance to therapy. By inhibiting RNR, BBI-825 deprives ecDNA-reliant cancer cells of dNTPs, leading to synthetic lethality in these tumor types. The drug has demonstrated preclinical efficacy in multiple tumor models and entered Phase 1/2 clinical trials (STARMAP study) focused on patients with solid tumors exhibiting resistance gene amplifications—particularly those with MAPK pathway activation such as BRAFV600E or KRASG12C mutations in colorectal cancer. However, following preliminary pharmacokinetic data from Part 1 of the trial showing a lack of dose-proportional exposure and increased development complexity/costs due to evolving treatment landscapes, Boundless Bio decided not to advance BBI-825 into further stages of clinical development[1][3][5][6][7].
