BBI-940 is a novel, orally bioavailable, highly selective kinesin degrader developed by Boundless Bio. It targets a previously undrugged kinesin protein that plays a critical role in DNA segregation during mitosis, including the segregation of extrachromosomal DNA (ecDNA), which is implicated in oncogene amplification and cancer progression. Preclinical studies have demonstrated potent anti-tumor activity for BBI-940 as a single agent, including tumor regression in mouse xenograft models of cancer. The drug represents the lead candidate from Boundless Bio’s Kinesin program and is positioned as a first-in-class therapy aimed at disrupting ecDNA-driven oncogenic processes in solid tumors[3][6][8].
