Overview

BBO-11818 + pembrolizumab is a combination therapy under clinical investigation for the treatment of advanced or metastatic solid tumors harboring KRAS mutations, such as non-small cell lung cancer, pancreatic ductal adenocarcinoma, and colorectal cancer. **BBO-11818** is an orally bioavailable small molecule dual inhibitor that non-covalently binds both the inactive GDP-bound (OFF) and active GTP-bound (ON) forms of KRAS. It exhibits potent inhibition of several cancer-driving KRAS mutants (including G12A, G12C, G12D, G12S, G12V) and is highly selective for KRAS over other RAS isoforms. **Pembrolizumab** is a monoclonal antibody targeting the immune checkpoint receptor PD-1. In the ongoing phase 1 KONQUER-101 trial, the combination is being assessed for safety, pharmacokinetics, and efficacy. The scientific rationale is that BBO-11818's direct KRAS inhibition may synergize with PD-1 blockade to augment anti-tumor immunity in KRAS-driven cancers[1][2][3][4][5][6].
