Overview

BBO-8520 is an investigational, orally bioavailable, small molecule covalent inhibitor of KRAS G12C. It is designed to bind and inhibit both the active (GTP-bound, ON) and inactive (GDP-bound, OFF) states of KRAS G12C. This dual inhibition provides optimal target coverage and addresses key mechanisms of adaptive resistance seen with current OFF-state inhibitors, such as KRAS G12C amplification and receptor tyrosine kinase activation. BBO-8520 has demonstrated substantial tumor growth inhibition in preclinical models, even after resistance to sotorasib (an FDA-approved OFF-state inhibitor). The drug is being developed for the treatment of cancers harboring KRAS G12C mutations, including non-small cell lung cancer (NSCLC), colorectal cancer (CRC), and potentially pancreatic cancer[2][6][8].
