Overview

BBP-398 + nivolumab is an investigational combination therapy for advanced solid tumors, particularly non-small cell lung cancer (NSCLC) with KRAS mutations. BBP-398 is a small molecule SHP2 inhibitor that blocks the activity of Tyrosine-protein phosphatase non-receptor type 11 (SHP2), a protein tyrosine phosphatase involved in the RTK-MAPK signaling pathway critical for cell growth and survival. By inhibiting SHP2, BBP-398 disrupts oncogenic signaling downstream of receptor tyrosine kinases and RAS/ERK MAPK pathways. Nivolumab (Opdivo) is a monoclonal antibody targeting Programmed cell death protein 1 (PD-1) on T cells to enhance anti-tumor immune responses. The combination aims to both inhibit tumor cell proliferation and boost immune-mediated tumor destruction. This regimen is being developed by BridgeBio Pharma/Navire Pharma in collaboration with Bristol Myers Squibb and LianBio for oncology indications including NSCLC with KRAS mutations[1][2][3][4][5][6][8].
