Drug intelligence / Profile preview
BBT-4437 is a novel, brain-penetrable, reversible pan-TEAD (TEA domain transcription factors 1-4) inhibitor that targets the Hippo signaling pathway [C133]. Developed by Bridge Biotherapeutics (now Parataxis Korea), BBT-4437 was designed to treat solid tumors, particularly those with aberrant Hippo signaling, such as NF2-deficient malignant mesothelioma [C133]. It acts by inhibiting TEAD transcriptional activity and blocking TEAD palmitoylation, which prevents the interaction of TEAD with the oncoproteins YAP and TAZ. Preclinical studies demonstrated that BBT-4437 inhibits cell proliferation in NF2-deficient mesothelioma cells and shows synergistic effects when combined with EGFR inhibitors like osimertinib or KRAS inhibitors like sotorasib. However, following the acquisition of Bridge Biotherapeutics by Parataxis Holdings and its subsequent rebranding to Parataxis Korea in August 2025, the company pivoted its business focus to digital asset treasury management, and development of BBT-4437 was discontinued.
