Overview

BBz CAR-T is a second-generation chimeric antigen receptor (CAR) T-cell therapy targeting the CD19 antigen, which is widely expressed on B-cell malignancies. Developed by Immunochina Pharmaceuticals, the therapy involves engineering a patient's T cells using a lentiviral vector to express a CAR construct consisting of an FMC63-derived CD19-specific single-chain variable fragment (scFv), a 4-1BB (CD137) co-stimulatory domain, and a CD3ζ intracellular signaling domain. In clinical studies for relapsed or refractory B-cell non-Hodgkin’s lymphoma (B-NHL), BBz CAR-T has demonstrated significant antitumor efficacy with a favorable safety profile. Compared to CD28-based CAR-T constructs (28z), the 4-1BB co-stimulatory domain in BBz CAR-T is associated with slower T-cell activation, enhanced persistence, and a lower incidence of severe cytokine release syndrome (CRS) and immune effector cell-associated neurotoxicity syndrome (ICANS).
