BC-3205 is a highly potent small-molecule antibiotic belonging to the pleuromutilin class, developed for the treatment of bacterial infections, particularly those caused by *Staphylococcus aureus*. It exerts its antibacterial effect by binding to the 50S ribosomal subunit at the peptidyl transferase center (PTC). The molecule's tricyclic mutilin core blocks the A-site, while its C14 extension points into the P-site, thereby perturbing tRNA accommodation and inhibiting bacterial protein synthesis. BC-3205 has demonstrated clinically relevant potency, often exceeding that of linezolid in preclinical models against *S. aureus*. It was developed by Nabriva Therapeutics as part of their advanced pleuromutilin platform.
