BC-DN-01 is a topical cream formulation of benfotiamine, a lipid-soluble derivative of thiamine (vitamin B1), developed by BioChemics, Inc. for the treatment of painful diabetic peripheral neuropathy (DPN). Benfotiamine acts as a prodrug that increases intracellular thiamine diphosphate levels, which in turn activates the enzyme transketolase. This activation helps divert glucose metabolites away from damaging biochemical pathways—such as the polyol, hexosamine, and protein kinase C pathways—and reduces the formation of advanced glycation end products (AGEs) and oxidative stress. By delivering benfotiamine topically, BC-DN-01 aims to provide localized therapeutic effects in the extremities of diabetic patients while minimizing systemic exposure and potential side effects.
