BC0001 is a preclinical-stage dual-payload antibody-drug conjugate (ADC) developed by Protheragen for the treatment of cancer. The therapeutic candidate specifically targets the epidermal growth factor receptor (EGFR), a transmembrane protein that is frequently overexpressed or mutated in various solid tumors, including lung, colorectal, and head and neck cancers. BC0001 utilizes a unique dual-payload architecture, where two distinct cytotoxic agents are conjugated to a single anti-EGFR monoclonal antibody. This approach is designed to enhance anti-tumor efficacy and potentially overcome drug resistance mechanisms that often limit the effectiveness of traditional single-payload ADCs. By binding to EGFR on the surface of malignant cells, the conjugate is internalized, leading to the intracellular release of the payloads and subsequent cell death.
