Overview

BC250 is a humanized bispecific T-cell engager (BiTE) designed to target CD19-expressing B-cell malignancies. Developed by researchers at Memorial Sloan Kettering Cancer Center, it utilizes an IgG(L)-scFv format to bridge CD19 on leukemia cells with CD3 on T cells, facilitating T-cell mediated cytotoxicity. In preclinical studies, BC250 demonstrated potent anti-leukemia activity against acute lymphoblastic leukemia (ALL) and Burkitt’s lymphoma, showing superior efficacy compared to the FDA-approved bispecific antibody blinatumomab in xenograft models. It has also been studied in combination with CD33-targeting bispecifics to address tumor heterogeneity in mixed phenotype acute leukemia.
