Overview

BC3195 is a first-in-class antibody-drug conjugate (ADC) targeting Cadherin 3 (CDH3) (P-cadherin), a calcium-dependent cell-cell adhesion glycoprotein highly expressed in various malignancies such as lung, breast, gastric, ovarian, colorectal, and pancreatic cancers. The ADC consists of an anti-CDH3 antibody linked via a cleavable dipeptide linker (Val-Cit) to the cytotoxic payload monomethyl auristatin E (MMAE), which disrupts microtubules. Upon binding to CDH3 on tumor cells, BC3195 is internalized and releases MMAE intracellularly to induce cell death; it also exerts a bystander effect on surrounding tumor cells. Preclinical studies demonstrated strong antitumor activity and favorable safety. Clinically, BC3195 has shown promising efficacy in advanced non-small cell lung cancer (NSCLC), especially in EGFR-mutant NSCLC patients. It is currently being evaluated in Phase 1 trials for multiple solid tumors including NSCLC, breast cancer, ovarian cancer, and other CDH3-expressing cancers[1][2][4][5][6][7].
