Overview

BC462 is a non-functional control bispecific T-cell engager (T-BsAb) used in preclinical research, specifically in the study of acute myeloid leukemia (AML). It is constructed using the IgG[L]-scFv 2+2 'da vinci' platform, which typically features two anti-CD3 arms attached to an IgG backbone. BC462 contains a mutated, non-binding sequence for the IL1RL1 (ST2) receptor, serving as a negative control for functional T-cell engagers like BC281. It allows researchers to account for non-specific effects of the antibody scaffold and T-cell engagement in immunocompetent murine models. The antibody features a silenced Fc domain to eliminate non-specific FcγR binding and complement activation, improving the accuracy of preclinical efficacy assessments.
