Overview

BC602 is an asymmetric bispecific antibody (BsAb) designed to target both leucine-rich repeat-containing G-protein coupled receptor 5 (LGR5) and epidermal growth factor receptor (EGFR). Developed by Dragon Boat Biopharmaceutical, the molecule features a VHH arm targeting LGR5 and a Fab arm targeting EGFR. It is engineered with an enhanced Fc region to increase FcγRIII binding and antibody-dependent cellular cytotoxicity (ADCC) activity. BC602 works by binding to both cell surface receptors, inducing significantly higher receptor internalization and degradation compared to monotherapy with either target-specific antibody alone. Preclinical studies in mouse CDX and PDX models have demonstrated superior anti-tumor efficacy in colorectal cancer (CRC) and head and neck squamous carcinoma (HNSCC) models. As of early 2026, the drug is in the IND-enabling stage with clinical trials planned for cancers overexpressing both targets.
