BC8-Fc-C825 is a bispecific recombinant fusion protein engineered for pretargeted radioimmunotherapy (PRIT) and designed specifically for the treatment of myeloid leukemia. It consists of an antibody fragment (BC8) targeting human CD45, a protein highly expressed on the surface of hematopoietic cells and myeloid leukemia cells, fused to an anti-yttrium-DOTA single-chain variable fragment (C825), which serves as a high-affinity capture domain for radiolabeled Y-DOTA-biotin. The mechanism allows for specific delivery of radiotherapy to leukemia cell sites: the fusion protein first binds to CD45-expressing tumor cells, and a radiolabeled ligand (such as ^90Y-DOTA-biotin) is administered later, selectively attaching to the tumor-localized fusion protein, thereby reducing off-target radiation exposure. The construct is under preclinical investigation as a novel approach to targeted radiotherapy for acute myeloid leukemia and related hematologic malignancies[1][3][9].
