Overview

**BCA356** is a preclinical, tumor-targeted immunocytokine developed by Bicara for CAIX-expressing solid tumors, notably clear cell renal cell carcinoma. It is a bispecific humanized IgG1-based fusion molecule that binds carbonic anhydrase IX and carries an attenuated interleukin-12 payload fused to its heavy chains in a knob-in-hole format. The design is intended to localize IL-12 activity to the tumor microenvironment, activate innate and adaptive antitumor immunity, retain antibody-dependent cellular cytotoxicity activity, and reduce the systemic toxicity associated with unconjugated IL-12.
