Overview

BCG + tremelimumab is an investigational combination therapy consisting of the live-attenuated bacterial immunotherapy Bacillus Calmette-Guérin (BCG) and the fully human monoclonal antibody tremelimumab (formerly CP-675,206). Tremelimumab is an immune checkpoint inhibitor that targets Cytotoxic T-Lymphocyte-Associated Antigen 4 (CTLA-4), thereby enhancing T-cell activation and proliferation. BCG is a standard intravesical treatment for bladder cancer that induces a local inflammatory and immune response. This combination was evaluated in a Phase I trial (NCT00880854) by the University of Wisconsin-Madison for patients with BCG-resistant localized transitional cell carcinoma of the bladder, aiming to overcome resistance by combining local and systemic immunotherapy. The trial was ultimately withdrawn due to poor accrual.
