BCG Glaxo is a specific substrain of the Bacillus Calmette-Guérin (BCG) vaccine, derived from a live attenuated strain of *Mycobacterium bovis*. It is classified as an evolutionarily "late" strain due to the loss of the Region of Differentiation 2 (RD2) genomic segment. Historically, it was utilized for the prevention of tuberculosis and as an intravesical immunotherapy for non-muscle invasive bladder cancer (NMIBC), particularly carcinoma in situ. The mechanism involves the stimulation of a local inflammatory and cellular immune response, characterized by the production of cytokines such as interleukin-6 and interleukin-8, which leads to the destruction of tumor cells. However, comparative studies have indicated that the Glaxo strain may be less immunogenic and efficacious than other substrains like Connaught or Russian. Production was eventually transferred to Evans Medical (later Medeva), and the strain is currently largely discontinued in clinical practice.
