Overview

**BCG-Moreau** is a live attenuated vaccine derived from the Mycobacterium bovis Bacillus Calmette–Guérin (BCG) strain, originally isolated in Rio de Janeiro, Brazil, and later produced in Poland and other regions. It is used for the prevention of **tuberculosis (TB)**. BCG-Moreau is distinguished genetically from its parental Brazilian substrain and other BCG substrains by unique deletions and polymorphisms—including the 975-bp deletion in the distal end of fadD26 and start of ppsA, IS6110 insertion in the phoP promoter, and a deletion within Rv3887c (membrane transport protein of the ESX-2 type VII secretion system). These genetic traits contribute to differences in immunogenicity and safety profile, with BCG-Moreau showing fewer adverse events (e.g., BCGosis) in immunocompromised populations compared to other BCG substrains. The vaccine works primarily through induction of cell-mediated immunity, activating T cells and particularly natural killer (NK) cells, which produce interferon-gamma (IFN-γ) crucial for mycobacterial defense[1][2]. Compared to other BCG strains, BCG-Moreau is defective in phthiocerol dimycocerosates (PDIMs) and phenolic glycolipids (PGLs), cell wall lipids important for virulence, resulting in greater attenuation and lower rate of complications[2].
