BCG010 is a fully human IgG1κ monoclonal antibody designed as a potentially best-in-class antagonist of the NKG2A (CD94-NKG2A) inhibitory checkpoint. Developed by Biocytogen in collaboration with Dragon Boat Biopharmaceutical, the antibody is engineered with an Fc LALA (L234A/L235A) substitution to eliminate Fc-mediated effector functions while maintaining high affinity and selectivity for human and cynomolgus NKG2A. By blocking the interaction between NKG2A on natural killer (NK) cells and CD8+ T cells and its ligand HLA-E on tumor cells, BCG010 alleviates inhibitory signaling and enhances anti-tumor cytotoxicity. Preclinical studies have demonstrated that BCG010 possesses superior affinity and a longer half-life compared to monalizumab analogs, with no cross-reactivity to the activating receptors NKG2C or NKG2E. It is currently in early development for the treatment of various cancers, showing efficacy in humanized mouse models both as a monotherapy and in combination with adoptive NK-cell therapy or PD-L1 inhibitors.
