BCG014 is a CDH3-targeting antibody-drug conjugate (ADC) being developed by Biocytogen for the treatment of various solid tumors. The drug consists of a fully human antibody (Ab.20), generated using the RenLite platform, which specifically binds to Cadherin-3 (CDH3/P-cadherin) with high affinity and exhibits robust internalization. This antibody is conjugated to BLD1102, a novel topoisomerase I (TOP1) inhibitor payload. CDH3 is frequently overexpressed in cancers such as pancreatic, head and neck, esophageal, breast, gastric, lung, and colorectal tumors, while showing limited expression in normal tissues. In preclinical studies, BCG014 has demonstrated significant anti-tumor activity in multiple patient-derived xenograft (PDX) models, outperforming benchmark conjugates.
