BCG015 is a first-in-class antibody-drug conjugate (ADC) targeting Transmembrane 4 L6 family member 5 (TM4SF5), developed by Biocytogen for the treatment of hepatocellular carcinoma (HCC). The drug consists of a high-affinity monoclonal antibody generated from the RenLite® transgenic mouse platform, conjugated to the BLD1102 linker-payload. TM4SF5 is a tumor-associated antigen highly expressed in approximately 70% of HCC cases with minimal expression in healthy tissues. Preclinical studies have demonstrated that BCG015 exhibits potent TM4SF5-dependent internalization and significant antitumor efficacy in various cell-derived and patient-derived xenograft models. As of early 2026, the program is undergoing preliminary safety evaluations in canine and cynomolgus monkey models.
