BCG016 is a novel bispecific antibody-drug conjugate (ADC) developed by Biocytogen that targets both trophoblast glycoprotein (5T4) and the membrane-bound C-terminal subunit of mucin 1 (MUC1-C). Utilizing the RenLite common light chain mouse platform, BCG016 is engineered to bind specifically to tumor cells co-expressing these two antigens, which are frequently overexpressed in a variety of solid tumors including lung, breast, and colorectal cancers. The ADC is conjugated to a topoisomerase I (TOP1) inhibitor payload via the BLD1102 linker-payload system. Preclinical evaluations in patient-derived xenograft (PDX) models have demonstrated that BCG016 exhibits superior anti-tumor efficacy compared to parental ADCs and benchmark treatments, suggesting its potential as a therapeutic candidate for cancers with high 5T4 and MUC1 expression.
