BCG017 is a first-in-class bispecific antibody-drug conjugate (ADC) designed to target both Protein Tyrosine Kinase 7 (PTK7) and Epidermal Growth Factor Receptor (EGFR). Developed using the RenLite common light chain mouse platform, the drug features a bispecific antibody backbone with a higher affinity for PTK7 than for EGFR, which is intended to enhance tumor selectivity and minimize on-target toxicity in normal tissues. BCG017 is conjugated to BLD1102, a proprietary topoisomerase 1 inhibitor (TOP1i) linker-payload. By dual-targeting these two antigens, BCG017 aims to overcome resistance mechanisms associated with traditional EGFR-targeted therapies and address tumor heterogeneity. Preclinical studies have demonstrated potent anti-tumor activity in various solid tumor models, including gastric, colorectal, esophageal, breast, and lung cancers.
