BCG018 is an antibody-drug conjugate (ADC) targeting integrin αvβ6 (ITGB6), a heterodimeric protein overexpressed in various solid tumors and associated with poor prognosis. Developed by Biocytogen using the RenLite® platform, BCG018 consists of a fully human monoclonal antibody (anti-ITGB6-B) conjugated to BLD1102, a novel topoisomerase I inhibitor payload. The antibody component selectively binds to ITGB6 and its heterodimer αvβ6 with high affinity and is effectively internalized into tumor cells without disrupting the interaction between ITGB6 and its natural ligand, LAP. In preclinical studies, BCG018 demonstrated superior anti-tumor efficacy compared to benchmark ADCs in patient-derived xenograft (PDX) and cell-derived xenograft (CDX) models of pancreatic cancer, colorectal cancer, and non-small cell lung cancer (NSCLC), regardless of ITGB6 expression levels.
