BCG022 is an AI-engineered, biparatopic bispecific T-cell engager (TCE) designed to target Delta-like ligand 3 (DLL3) and CD3. Developed by Biocytogen using its proprietary RenMice (RenLite) platform, the molecule binds to two distinct, non-overlapping epitopes on the DLL3 antigen (biparatopic binding) while simultaneously engaging the CD3 epsilon subunit on T cells. This dual-epitope targeting of DLL3 is intended to enhance binding specificity and tumor-cell killing potency, particularly in tumors with low or heterogeneous DLL3 expression, while the optimized CD3 affinity aims to minimize systemic cytokine release syndrome (CRS). Preclinical studies in small cell lung cancer (SCLC) and neuroendocrine carcinoma (NEC) models have demonstrated superior anti-tumor activity compared to tarlatamab analogs, with a favorable safety profile and low cytokine release observed in non-human primates.
