BCG022-vcMMAE is a novel bispecific antibody-drug conjugate (bsADC) developed by Biocytogen Pharmaceuticals. It is composed of a fully human bispecific antibody, BCG022, which targets both the Human Epidermal Growth Factor Receptor 3 (HER3) and the Mesenchymal-epithelial transition factor (MET). The antibody is conjugated to the cytotoxic payload monomethyl auristatin E (MMAE) via a protease-cleavable valine-citrulline (vc) linker. HER3 and MET are frequently co-expressed in various malignancies, including non-small cell lung cancer (NSCLC), gastric, colorectal, and breast cancers, where they often contribute to therapeutic resistance, particularly against EGFR tyrosine kinase inhibitors. By simultaneously binding to both targets, BCG022-vcMMAE facilitates enhanced receptor internalization and delivers the microtubule-disrupting payload directly into tumor cells. Preclinical evaluations in cell-derived and patient-derived xenograft models have demonstrated potent anti-tumor activity across multiple cancer types.
