Overview

BCG024 is a novel trispecific T-cell engager (TriAb) targeting DLL3, CD3, and 4-1BB, developed by Biocytogen. It is designed to treat DLL3-positive solid tumors, specifically small cell lung cancer (SCLC). The molecule utilizes a VHH-based 4-1BB agonist derived from the RenNano platform to provide tumor-antigen-dependent co-stimulation. This approach aims to overcome the limitations of conventional T-cell engagers, such as T-cell exhaustion, by enhancing T-cell persistence and functionality. Preclinical studies have demonstrated that BCG024 exhibits robust anti-tumor efficacy in SCLC xenograft models and maintains a favorable safety profile with minimal systemic cytokine release.
