BCG026 is a novel bispecific antibody-drug conjugate (bsADC) developed by Biocytogen, designed to target both the tumor microenvironment and the tumor cells themselves. It specifically binds to Fibroblast Activation Protein (FAP), which is highly expressed on cancer-associated fibroblasts (CAFs) in the stroma, and Glypican 1 (GPC1), a cell surface protein overexpressed in various solid tumors, particularly pancreatic adenocarcinoma (PAAD). By targeting CAFs, BCG026 aims to deplete the dense fibrotic stroma that often hinders drug delivery and promotes resistance. The molecule features a fully human bispecific antibody backbone generated via the RenLite platform, conjugated to a topoisomerase I inhibitor (TOP1i) payload using the BLD1102 linker-payload system. Preclinical data indicate that BCG026 exhibits potent anti-tumor activity and a CAF-dependent bystander killing effect, making it a promising candidate for treating PAAD, lung cancer, and other stroma-rich solid tumors.
