BCG029 is an antibody-drug conjugate (ADC) designed to target A disintegrin and metalloproteinase domain-containing protein 9 (ADAM9), a membrane-anchored protein overexpressed in various solid tumors including pancreatic, esophageal, breast, gastric, lung, and colorectal cancers. Developed by Biocytogen using their proprietary RenLite ADAM9 knockout mouse platform, BCG029 utilizes a fully human monoclonal antibody (clone Ab.01) that specifically binds to the membrane-proximal region of the ADAM9-L isoform while avoiding the secreted ADAM9-S isoform. This targeting strategy is intended to improve specificity and reduce off-target toxicity, particularly by minimizing binding to myeloid cells such as monocytes and neutrophils. The antibody is conjugated to BLD1102, a novel topoisomerase I (Top1) inhibitor payload. In preclinical studies using patient-derived xenograft (PDX) models, BCG029 has demonstrated potent anti-tumor efficacy across multiple cancer types.
