BCG033 is a first-in-class bispecific antibody-drug conjugate (ADC) developed by Biocytogen Pharmaceuticals that simultaneously targets two tumor-associated antigens: protein tyrosine kinase 7 (PTK7) and trophoblast cell-surface antigen 2 (TROP2). Generated using the proprietary RenLite® platform, BCG033 features a fully human bispecific antibody with a common light chain, conjugated to the cytotoxic payload monomethyl auristatin E (MMAE) via the proprietary BLD1102 linker. This dual-targeting approach is designed to enhance therapeutic efficacy and selectivity while reducing off-target effects compared to conventional single-target ADCs. Preclinical studies have demonstrated significant anti-tumor activity in models of triple-negative breast cancer (TNBC), non-small cell lung cancer (NSCLC), gastric cancer, and colorectal cancer. The drug is currently in the preclinical stage of development.
