Overview

BCG037 is a first-in-class, fully human IgG1 antibody-drug conjugate (ADC) targeting placental alkaline phosphatase (ALPP) and germ cell alkaline phosphatase (ALPG). Developed by Biocytogen using their RenMab™ humanized mouse platform, the antibody component exhibits high affinity and cross-reactivity for both human and cynomolgus monkey ALPP/ALPG, while avoiding other alkaline phosphatase family members like ALPL and ALPI. The ADC is conjugated to a potent topoisomerase I (Top1) inhibitor payload with a high drug-to-antibody ratio (DAR) of approximately 8. In preclinical studies, BCG037 demonstrated significant tumor growth inhibition in pancreatic and gastric cancer patient-derived xenograft (PDX) models, outperforming benchmark ADCs without treatment-related toxicity.
