BCG039

BCG039 is a bispecific antibody-drug conjugate (bsADC) developed by Biocytogen that simultaneously targets Nectin-4 and TROP2. Both targets are clinically validated in urothelial carcinoma (UC), with Nectin-4 targeted by enfortumab vedotin and TROP2 by sacituzumab govitecan. BCG039 utilizes a fully human bispecific antibody generated via the RenLite transgenic mouse platform. It is conjugated to BLD1102, a proprietary linker-payload system consisting of a novel topoisomerase 1 (TOP1) inhibitor and a highly hydrophilic, enzyme-cleavable linker. Preclinical studies in CDX and PDX models of urothelial carcinoma and other solid tumors have demonstrated superior anti-tumor activity compared to single-targeting parental ADCs and benchmark agents, potentially offering improved efficacy and reduced toxicity through cooperative binding of the two antibody arms.
