BCG044

BCG044 is a novel bispecific antibody-drug conjugate (ADC) designed to target both Epithelial Cell Adhesion Molecule (EpCAM) and Human Epidermal Growth Factor Receptor 3 (HER3). Developed by Biocytogen, the drug utilizes a "1+1" bispecific antibody format to enhance tumor selectivity and maximize receptor-mediated internalization while mitigating the systemic toxicities, such as gastrointestinal issues and pancreatitis, often associated with monospecific EpCAM-targeting agents. BCG044 is conjugated to BLD1102, a proprietary topoisomerase I (TOP1) inhibitor linker-payload. Preclinical studies have demonstrated that simultaneous engagement of both EpCAM and HER3 by BCG044 leads to superior internalization and potent anti-tumor efficacy in various solid tumor models, including colorectal, breast, and non-small cell lung cancer (NSCLC), with an improved safety profile compared to benchmark ADCs.
