BCG045 is a first-in-class bispecific antibody-drug conjugate (ADC) designed to simultaneously target Trophoblast cell-surface antigen 2 (TROP2) and Mucin 1 (MUC1). Developed by Biocytogen, the drug is engineered to address common challenges associated with TROP2-targeted therapies, such as on-target toxicity and resistance. The bispecific antibody component utilizes a low-affinity TROP2 binder to minimize internalization in normal tissues and a MUC1 binder that specifically recognizes the MUC1-N/C junction of membrane-bound MUC1, thereby avoiding neutralization by shed MUC1 in circulation. BCG045 is conjugated to BLD1102, a topoisomerase I inhibitor (TOP1i) linker-payload. Preclinical studies presented at AACR 2026 demonstrated that BCG045 has broad binding capabilities across various tumor cell lines and exhibits potent anti-tumor efficacy in multiple patient-derived xenograft (PDX) models of epithelial tumors, showing a potentially improved safety and efficacy profile compared to benchmark ADCs.
