BCG046 is an antibody-drug conjugate (ADC) targeting the C-terminal fragment (CTF) of CUB domain-containing protein 1 (CDCP1). Developed by Biocytogen using their proprietary RenMice platform, the candidate comprises a fully human monoclonal antibody (clone Ab.168) conjugated to the microtubule-disrupting agent monomethyl auristatin E (MMAE) via a protease-cleavable valine-citrulline (vc) linker. CDCP1 is a transmembrane glycoprotein frequently overexpressed in various malignancies, including colorectal, lung, and pancreatic cancers, where it correlates with tumor progression and poor prognosis. By specifically targeting the CTF, BCG046 is designed to maintain efficacy even in the presence of shed amino-terminal fragments (ATF) in the systemic circulation. Preclinical data demonstrate that BCG046 exhibits potent internalization and significant anti-tumor activity in patient-derived xenograft (PDX) models of colorectal cancer and non-small cell lung cancer (NSCLC).
