BCG047 is a novel heavy-chain antibody-drug conjugate (HcADC) targeting Folate Receptor 1 (FOLR1, also known as FRα). It is composed of a fully human single-domain antibody (VHH), HCAb-01, which was identified using the RenNano mouse platform, conjugated to BLD1102, a topoisomerase I inhibitor (TOP1i) linker-payload. BCG047 is designed to target tumor cells with a wide range of FOLR1 expression levels, including those with low expression that are typically underserved by first-generation FRα-targeted ADCs like mirvetuximab soravtansine. Preclinical data indicate that BCG047 exhibits potent antitumor activity in patient-derived xenograft (PDX) models and possesses favorable pharmacokinetic and safety profiles.
