Overview

BCG@PLTm is an autonomously propelled, urea-powered platelet-derived nanomotor (PLTm) encapsulating Bacillus Calmette-Guérin (BCG), developed for the treatment of immunotherapy-refractory bladder cancer. The nanomotor utilizes the decomposition of urea in urine to drive active penetration through the dense extracellular matrix (ECM) of bladder tumors, overcoming the stromal barrier that typically limits intravesical BCG therapy. Mechanistically, BCG@PLTm restores the expression of death-associated protein like 1 (DAPL1), which reverses pathological ECM stiffening and alleviates immune exclusion, thereby reactivating the infiltration of cytotoxic T-cells. Preclinical studies in orthotopic mouse models have demonstrated significant tumor growth inhibition (rate > 90%) and a favorable safety profile.
