**bcl-2 siRNA** is a synthetic small interfering RNA specifically designed to silence the BCL2 gene, which encodes B-cell lymphoma 2, a protein that inhibits apoptosis and promotes tumor survival. This siRNA acts by RNA interference, binding to BCL2 mRNA and promoting its degradation, leading to reduced BCL2 protein expression. In preclinical studies, bcl-2 siRNA has been delivered using various nanoparticle formulations (including liposomal and polymer-based systems), resulting in effective silencing of BCL2, antitumor effects, induction of apoptosis, and chemosensitization across multiple cancer models including breast, pancreatic, bladder, and hematologic cancers. The main mode of action is as an antineoplastic agent via RNA-mediated knockdown of an antiapoptotic factor, and it is under investigation as both monotherapy and in combination with chemotherapy[1][4][6][7].
