Overview

BCL7A shRNA is a short hairpin RNA (shRNA) research tool designed to silence the expression of the BCL7A gene through RNA interference. BCL7A is a member of the canonical m-SWI/SNF (BAF) chromatin remodeling complex and is frequently lost in primary multiple myeloma (MM) cells. In preclinical research, BCL7A shRNA is used to model this loss, which results in increased DNA accessibility at specific genomic regions and the activation of the IRF4 oncoprotein. The knockdown of BCL7A leads to enhanced IRF4-dependent transcriptional activity, promoting a more proliferative phenotype in myeloma cells. This tool is primarily utilized in in-vitro and in-vivo models to investigate the molecular mechanisms of myeloma pathogenesis and the development of IRF4 dependency.
