Allogene's **BCMA/CD70 dual CAR T-cell construct** is a preclinical, off-the-shelf allogeneic CAR-T approach for high-risk multiple myeloma. Healthy-donor T cells are CRISPR edited to disrupt the **TRAC** locus and enable site-specific integration of bicistronic CARs against B-cell maturation antigen and CD70. The BCMA CAR directs cytotoxicity toward BCMA-expressing myeloma cells, while the CD70 CAR is intended both to target CD70-positive myeloma cells and to eliminate CD70-positive alloreactive T cells that could reject the donor-derived product. In preclinical studies, the dual-targeted cells showed activity against BCMA-loss antigen-escape models, resistance to allorejection, repeated-stimulation killing, and xenograft activity.
