Overview

BCMA BBz CAR refers to a chimeric antigen receptor (CAR) T-cell therapy architecture designed to target the B-cell maturation antigen (BCMA), a protein predominantly expressed on the surface of malignant plasma cells in multiple myeloma. The 'BBz' nomenclature specifies the intracellular signaling components: a 4-1BB (CD137) costimulatory domain and a CD3-zeta (CD3ζ) primary signaling domain. This specific configuration is widely utilized in clinical development due to the 4-1BB domain's ability to enhance T-cell metabolic fitness and promote long-term persistence in vivo compared to first-generation or CD28-based CARs. In the context of current research, BCMA BBz CARs serve as a standard-of-care benchmark for evaluating novel therapeutic formats, such as multispecific hybrid T-cell receptors (TCR/CARs), which aim to overcome antigen heterogeneity and prevent tumor relapse.
