BCMA BCL2L1 CAR is an experimental, 'armored' second-generation chimeric antigen receptor (CAR) T-cell therapy designed for the treatment of relapsed or refractory multiple myeloma. The construct utilizes a lentiviral vector to co-express an anti-BCMA scFv-41BBz CAR alongside BCL2L1 cDNA, which encodes the anti-apoptotic protein BCL-XL. This armoring strategy is intended to enhance CAR T-cell survival, proliferation, and persistence by providing resistance to activation-induced cell death (AICD) and overcoming exhaustion typically seen in conventional BCMA-targeted therapies. Preclinical data presented by researchers at the University of Calgary demonstrate that this construct maintains superior cytolytic activity and tumor clearance in multiple myeloma models, particularly those expressing the FAS death receptor ligand.
