BCMA-BiTE

BCMA-BiTE (B-cell Maturation Antigen Bispecific T-cell Engager) refers to a class of bispecific antibodies, primarily developed using Amgen's proprietary BiTE platform, designed to treat multiple myeloma. These molecules are engineered to simultaneously bind to BCMA, a protein highly expressed on the surface of malignant plasma cells, and the CD3 epsilon subunit of the T-cell receptor complex on T cells. This dual binding brings T cells into close proximity with tumor cells, bypassing the need for MHC-I-restricted antigen presentation and triggering T-cell activation, cytokine release, and perforin-mediated lysis of the cancer cells. Notable examples of BCMA-BiTE candidates include pavurutamab (AMG 701) and AMG 420.
