Overview

**BCMA-BTZ-NPs** is a preclinical BCMA-targeted bortezomib nanotherapy for multiple myeloma. It encapsulates the proteasome inhibitor bortezomib in PEG-PLGA nanoparticles functionalized with antibodies against B-cell maturation antigen, enabling selective uptake by BCMA-expressing myeloma cells. In preclinical multiple-myeloma models, the formulation increased tumor-cell internalization, proteasome inhibition, apoptosis, immunogenic cell death, antitumor immune activation, tumor accumulation, and survival relative to free bortezomib or non-targeted bortezomib nanoparticles. Receptor-mediated uptake may also help circumvent P-glycoprotein-associated bortezomib resistance while reducing off-target toxicity.
