Overview

BCMA CAR-CXCR4 NK cells are an investigational cell therapy product engineered from natural killer (NK) cells to express two functional transgenes: a chimeric antigen receptor (CAR) specific for B-cell maturation antigen (BCMA) and the chemokine receptor CXCR4. The CAR targets BCMA, which is highly expressed on multiple myeloma cells, allowing the NK cells to selectively recognize and kill these tumor cells. The enforced expression of CXCR4 enhances the migration of these NK cells toward regions producing the chemokine CXCL12, such as the bone marrow niche, where multiple myeloma typically resides. Mechanistically, co-expression of CXCR4 not only boosts tumor-directed trafficking, but also augments antigen-dependent cytotoxicity and signal transduction via increased ZAP-70 recruitment and ERK1/2 activation, thus intensifying and accelerating the killing of BCMA-expressing tumor cells both in vitro and in preclinical models. This therapeutic approach is being investigated primarily for multiple myeloma[1][2][3][4].
