BCMA CAR gamma-delta (γδ) T cells are an investigational allogeneic cell therapy platform designed to target B-cell maturation antigen (BCMA), a protein highly expressed on malignant plasma cells in multiple myeloma. Unlike conventional alpha-beta (αβ) T cells, γδ T cells recognize targets in an MHC-independent manner, which significantly reduces the risk of graft-versus-host disease (GvHD) and facilitates the development of "off-the-shelf" standardized therapies. As demonstrated by Unicet Biotech's preclinical candidate, these cells can be engineered with a third-generation CAR construct for dual-targeting (e.g., CD19 and BCMA) to mitigate antigen escape and improve the depletion of pathogenic B cells and long-lived plasma cells. Furthermore, the incorporation of an intracellular IL-15 (icIL-15) pro-survival module enhances the functional persistence and serial killing capacity of the cells in cytokine-free environments. This platform is being explored for the treatment of hematologic malignancies, such as multiple myeloma and B-cell acute lymphoblastic leukemia, as well as B-cell mediated autoimmune diseases.
