Overview

BCMA CAR-iNKT is an investigational **allogeneic cell therapy** in which human invariant natural killer T cells (iNKT) are engineered to express a chimeric antigen receptor (CAR) targeting **B cell maturation antigen (BCMA)**, a receptor highly expressed on malignant plasma cells in multiple myeloma. This approach combines the innate anti-tumor activity, tissue homing, and immune regulatory properties of iNKT cells with the tumor-targeting specificity of CARs. iNKT cells do not cause graft-versus-host disease, enabling the creation of "off-the-shelf" allogeneic products with superior cell fitness, reduced treatment time, and lower cost compared to autologous CAR-T therapies. Preclinical studies demonstrate anti-myeloma efficacy and suggest a potentially favorable safety profile with a lower or intermediate risk of cytokine release syndrome. In vivo persistence and efficacy have been improved by armoring with long-acting IL-7 (NT-I7). Early trials for multiple myeloma are advancing toward phase 1 clinical testing[1][6][7][3][4].
