BCMA CAR-NK (PRG-NK01) is an allogeneic, "off-the-shelf" chimeric antigen receptor natural killer (CAR-NK) cell therapy developed by Shenzhen Pregene Biopharma for the treatment of relapsed/refractory multiple myeloma. The therapy consists of NK cells derived from healthy donor peripheral blood, genetically engineered via lentiviral transduction to express a CAR targeting B-cell maturation antigen (BCMA). The specific construct utilizes a single-domain antibody (VHH/nanobody) for BCMA recognition, a CD28 costimulatory domain, and ectopic expression of interleukin-15 (IL-15) to promote NK cell persistence and anti-tumor activity. By using NK cells instead of T cells, the therapy aims to reduce the risk of cytokine release syndrome (CRS) and graft-versus-host disease (GvHD) while providing a more accessible treatment option.
