BCMA CAR T BCLxL Δ46-83 is a preclinical, research-stage armored chimeric antigen receptor (CAR) T-cell therapy designed for the treatment of multiple myeloma. The construct consists of autologous T cells engineered to express a CAR targeting B-cell maturation antigen (BCMA/CD269) and co-express a mutant form of the anti-apoptotic protein BCLxL (encoded by BCL2L1) featuring a deletion in the regulatory loop domain (Δ46-83). This modification is intended to enhance the survival and metabolic fitness of the CAR T cells, preventing progressive exhaustion and maintaining a central memory (TCM) phenotype under chronic antigenic stimulation and hypoxic conditions. Research presented at ASH 2023 demonstrated that this armored construct provides superior tumor clearance and persistence compared to unarmored BCMA CAR T cells in xenograft models.
